Depends what you want to do.

If just to backup the file (to keep safe) - yes. Use the backup option in PC Connect or the on-board backup option in the menory card option (Extras / Memory Card / Options / Backup). I prefer the second option as it means I have the backup with me (just in case) and it zips up the file quite well too, so it does not take up too much space. Both of these do backup ALL files, not just SMS, so may take a few minutes.

If you want to view them on the PC and reply to them or write new ones, then no, that is not possible, which is a shame - I would like that option too!

However, as boogie says in the message below, there are commercial programs available that may allow this. I just haven't tested them (yet).
