I suggest you please go to
https://www.symbiansigned.com/app/page/devcertgeneral and read all the
documents!
DevCertTool can make a .cer and a .key and a .csr (or you can input existing
ones). The .key file is what you use in signsis. The .csr is what you send
to SymbianSigned, and the .cer file can be thrown away.
SymbianSigned give you back a new .cer file ... you use this and the
original .key to do the signing.
