I have recently obtained a Nokia 9500 on Ebay for good price. How do I update the firmware to 6.27? Do I have to flash it with a specific application?

If not, then visit your nearest Nokia authorized service/repair center.

I am looking to flash it. There are no service centres where I am and NSU will not update it.
The firmware version on my 9500 is 4.37, the latest is 6.27.

In that case, I don't know; you probably would need to resort illegally distributed copyrighted tools, possibly also special hardware (a flashing station/cables) as the phone is an older model where it might not be possible via USB, and illegally distributed firmware image files.
If there's no Nokia service center nearby, find the nearest one and ask them whether you can send the phone there.
